Abstract

Background: This study focused on the effect of two screening and cultural methods on morphology and differentiation of result cells. Objective: Screening of chicken bone marrow mesenchymal stem cells with two different methods, direct and Ficol mounted and then their impact on morphology and differentiation of the cells. Methods: In this experimental study which was done in Pastore institute in 2009, 1 mg of 2 weeks old Raf chicken aspirated bone marrow of tibia, cultured within low glucose DMEM, 10% FBS, penicillin and streptomycin, with both direct (red blood cells remained) and Ficol (for red blood cells screening) mounted culture methods. After 4 passages, bone marrow cells of two cell culture methods were compared for their morphology and differentiation to bone, cartilage and adipose. Data were analysed by ANOVA test. Findings: In direct cell culture more colonization took place in comparing with Ficol mounting method and most cells had fibroblastic morphology. Results showed higher yields of Differentiation to bone, cartilage and adipose in direct method, respectively 21, 27 and 40%. Conclusion: Results indicated that screening and proliferation of mesenchymal stem cells with direct culturing system might be better way to screen chicken bone marrow mesenchymal stem cells than Ficol mounting method.
